Absence of fluorescence quenching in a photosynthetic mutant of Euglena gracilis.

چکیده
Substantial experimental evidence indicates that the level of chlorophyll fluorescence in algae and higher plants is controlled predominantly by the activity of photosystem II (PS II) of the photosynthetic electron transport chain (2). The overall fluorescence yield seems to depend on the oxidation/reduction state of Q, the presumed primary electron acceptor of PS II (7) (H2O ? Y hv hv -> Q -) A -* ? -> P700 XH). AcPS II PS I cording to Duysens' hypothesis (4), the oxidized form of Q quenches chlorophyll fluorescence, while the reduced form (QH) does not. We report here a photosynthetically defici.ent strain of Eutglena which appears to be lacking the fluorescence quenching normally associated with the {presence of Q. Eiglena. gracilis (strain Z) was cultured as previously described (9). The pale green mutant P4 is unable to carry out photosynthetic electron transport because of a block at or near light reaction II; detailed studies of this strain have been reported elsewhere (10). Chloroplast fragments were prepared according to Katoh and San Pietro (6). Fluorescence was measured with the technique devised by Duysens (3). Whole cells or chloroplasts were illuminated with a modulated monochromatic light beam (X = 435 nm, 270 cps), and the fluorescence emis;sion was measured with a photomultiplier/amplifier system tuned to the chopping frequency. Chromium- and copper-induced inhibition of photosynthesis in Euglena gracilis analysed on the single-cell level by fluorescence kinetic microscopy.
Here, we investigated effects of copper (Cu) and chromium (Cr) toxicity on two contrasting strains of Euglena gracilis, with and without chloroplasts, grown in culture media promoting either phototrophic or heterotrophic growth. This led to insights into Cr/Cu toxicity mechanisms and into the regulation of phototrophic vs heterotrophic metabolism. Our data strongly suggest that in Cu(2+) and Cr...
متن کاملThe Carotenoid Hydrocarbons of Euglena gracilis and Derived Mutants.
An examination of the carotene fractions extracted from Euglena gracilis Z and pressure-bleached Euglena mutants PR-1, PR-2, PR-3, and PR-4 revealed phytoene in mutants PR-1, PR-2, and PR-3. Photosynthetic E. gracilis Z cultured at different light intensities showed no detectable phytoene, nor was phytoene found in mutant PR-4. However, dark-cultured E. gracilis Z yielded readily assayable amou...
متن کاملEvolution of the chloroplast genome in photosynthetic euglenoids: a comparison of Eutreptia viridis and Euglena gracilis (Euglenophyta).
The chloroplast genome of Eutreptia viridis Perty, a basal taxon in the photosynthetic euglenoid lineage, was sequenced and compared with that of Euglena gracilis Ehrenberg, a crown species. Several common gene clusters were identified and gene order, conservation, and sequence similarity was assessed through comparisons with Euglena gracilis. Significant gene rearrangements were present betwee...
متن کاملChlorophyll Formation and Photosynthetic Competence in Euglena During Light-Induced Chloroplast Development in the Presence of 3, (3,4-dichlorophenyl) 1,1-Dimethyl Urea (DCMU).
The possibility that photosynthetic competence is gratuitous for light-induced chloroplast development in Euglena gracilis var. bacillaris was examined by incubating dark-grown resting cells in the light with DCMU, an inhibitor of photosynthesis. Under these conditions photosynthetic carbon dioxide fixation was inhibited essentially completely at all times during chloroplast development, but ab...
متن کاملGamma-irradiation effect: variation of photosynthetic activity of Euglena.
OBJECTIVE To study the effects of gamma-ray irradiation on carbon fixation (Specific production rate: SPR), CO2 utilization efficiency (CUE) and electron transfer rate (ETR) in the photosynthetic flagellate Euglena gracilis strain Z in a dose-response dependent manner. METHODS Euglena cells were cultured in an inorganic nutrient medium containing ammonium chloride or proteose peptone. Cells w...
